As a drug enters the body, its dynamics and metabolism can affect its final effects and even induce off-target effects that impact tissues and organs. To model drug dynamics in the microenvironment, OrganRXTM is used with multiple cell types forming a multi-organ system. As specific organs such as the liver or gut metabolize a drug, the effects of the metabolites on the other organs can be observed as modeled on the OrganRXTM system. Liquid chromatography-mass spectroscopy (LC-MS) is an analytical tool used to measure drug metabolism, offering a more detailed analysis of the effects of drugs on the organ system.
LC-MS characterizes biomarkers from the cell media before and after applying a drug to reveal the metabolites of the drug produced by specific tissues. LC-MS is advantageous for identifying metabolites at high sensitivity down to nanomolar and high-throughput analysis.
Currently, LC-MS analysis of drug metabolism in OrganRXTM is done by removing cell media and analyzing the drug consumption and production of metabolites by each organ. LC-MS sampling with OrganRXTM can provide faster and more accurate results for high throughput assays such as toxicity assays.
