TUMOR MICROENVIRONMENT FOR THERAPEUTIC SCREENING

Using the OrganRX™ Plate and OrganRX™ System, cell-cell and cell-matrix interactions within 3D tumor spheroid models is studied to provide deeper insights into drug response and tumor behavior. The platform is designed to accelerate cancer drug discovery and improve predictability of therapies using high-throughput & non-invasive imaging, cell viability assays, cell migration assays, flow cytometry, and cytokine release assays. The OrganRX™ Plate can support a coculture of tumor spheroids, stromal, endothelium, and immune cells, effortlessly creating a dynamic and physiologically relevant tumor microenvironment

Immunoncology (IO) cells are introduced into the outer wells, while tumor spheroids/organoids reside in adjacent stromal/tumor chambers. We can collect the flowing IO cells and measure activation markers by flow cytometry or endpoint immunofluorescence on the cancer cells after recirculation. Functional readouts of cytotoxicity include tumor cell viability assays (ATP/LDH, Calcein/EthD-III), ELISA-based cytokines/chemokines assays and live-cell imaging of tumor spheroids to track apoptosis and infiltration.
